A domain containing the ESR motif in ENHANCER OF SHOOT REGENERATION 1 functions as a transactivation domain

چکیده
The Arabidopsis ENHANCER OF SHOOT REGENERATION 1 (ESR1) gene is thought to regulate the initiation of in vitro shoot regeneration. ESR1 encodes a transcriptional activator belonging to the ETHYLENE-RESPONSIVE FACTOR family. In this study, we investigated the functions of the C-terminal domain of the ESR1 protein. Effectorreporter assays using the GAL4 DNA-binding domain fused with portions of the C-terminal domain revealed that the ESR motif, which is a short amino acid sequence commonly seen in the C-terminal domain of ESR1 and ESR2, functions as a transactivation domain. We also examined the effects of partial deletions of the C-terminal domain on the in vitro shoot regeneration-enhancing effects of ESR1 overexpression. The results revealed that the ESR motif and a region between the AP2/ERF domain and the ESR motif were required for the shoot regeneration-enhancing effects of ESR1. The region between the AP2/ERF domain and the ESR motif appeared to have an essential function other than nuclear localization, DNA-binding, and transactivation. Substitution of the ESR motif by the VP16 transactivation domain abolished the ability of ESR1 to enhance shoot regeneration, suggesting that the ESR motif not only has transactivation activity but also has other functions.

CCAAT/enhancer-binding proteins (C/EBPs) are transcriptional regulators implicated in cell proliferation, differentiation, survival, and tumorigenesis. Their biological activities require interactions with several protein partners. This report presents insights from in silico analysis aimed at identifying phosphorylation-dependent protein recognition motifs in C/EBPs. (1) All C/EBP variants con...
متن کاملPositive and negative regulation of the composite octamer motif of the interleukin 2 enhancer by AP-1, Oct-2, and retinoic acid receptor
The differentiating agent retinoic acid (RA) has been previously reported to interfere with 12-O-tetradecanoyl-phorbol-13-acetate (TPA)/Ca2+-induced signals for the regulation of the -96 to -66-bp octamer motif found in the enhancer for the interleukin (IL)-2 gene, which encodes a major T lymphocyte growth factor. The IL-2 octamer motif is a composite cis-element which binds Oct-1 and Oct-2 as ...

The magnetoresistance of a one-dimensional electron gas in a metallic ferromagnetic nanowire containing two atomic-size domain walls has been investigated in the presence of spin-orbit interaction. The magnetoresistance is calculated in the ballistic regime, within the Landauer-Büttiker formalism. It has been demonstrated that the conductance of a magnetic nanowire with double domain walls...
متن کاملDesigning Of Degenerate Primers-Based Polymerase Chain Reaction (PCR) For Amplification Of WD40 Repeat-Containing Proteins Using Local Allignment Search Method
Degenerate primers-based polymerase chain reaction (PCR) are commonly used for isolation of unidentified gene sequences in related organisms. For designing the degenerate primers, we propose the use of local alignment search method for searching the conserved regions long enough to design an acceptable primer pair. To test this method, a WD40 repeat-containing domain protein from Beauveria bass...

The human splicing factor ASF/SF2 (alternative splicing factor/splicing factor 2) is modular in structure with two RNA-binding domains (RBD1 and RBD2) and a C-terminal domain rich in arginine-serine dipeptide repeats. ASF/SF2 is an essential splicing factor that also functions as an important regulator of alternative splicing. In adenovirus E1A (early region 1A) alternative pre-mRNA splicing, A...
